Mechanistic impact of fibrosis by parathyroid hormone excess

چکیده

The fibrotic impact of parathyroid hormone (PTH) excess is an important aspect primary hyperparathyroidism (PHPT) that can contribute to organ dysfunction and morbidity. Understanding the underlying mechanisms PTH-induced fibrosis identifying potential therapeutic targets may pave way for novel treatment strategies. Further research needed elucidate complex interactions between PTH pathways evaluate efficacy targeted interventions in preventing or reversing associated with PHPT. observed across multiple systems. Fibrosis resulting from impair function lead significant morbidity mortality. involved crucial developing therapies mitigate its detrimental effects.

Parathyroid hormone without parathyroid glands.

In mammals, PTH is a component in a complex signaling system involving three structurally related ligands and two G protein-coupled receptors, which share significant amino acid sequence homology (Fig. 1). PTH is a peptide hormone comprising 84 amino acids that is made by the parathyroid glands (and to a much smaller extend by the thymus) and acts through the PTH/PTH-related peptide (PTHrP) rec...

Stimulation of Enzyme Activity by Parathyroid Hormone

Received for publication 26 October 1976 and in revised form 24 January 1977. INTRODUCTION The fat cell adenylate cyclase is a plasma membrane-bound enzyme which is supposed to play a central role in the stimulation of lipolysis by hormones (1). The rat fat cell enzyme reacts to a variety of hormones including catecholamines and peptide hormones such as ACTH, glucagon, and secretin (2-4). By co...

Conversion of proparathyroid hormone to parathyroid hormone by a particulate enzyme of the parathyroid gland.

The conversion of proparathyroid hormone (proparathormone) to parathyroid hormone (parathormone) by subcellular fractions of the bovine parathyroid has been investigated. The identification of the conversion product as parathormone was established by its elution postion during ion exchange chromatography and gel filtration, and by partial amino acid sequence analysis of its NH2-terminal region....

Biosynthesis of proparathyroid hormone and parathyroid hormone by human parathyroid glands.

Human parathyroid glands obtained at autopsy were incubated with [(3)H]leucine and [(3)H]lysine. After incubation, nonradioactive parathyroid tissue of either human or bovine origin was added. Radioactive parathyroid hormone and proparathyroid hormone were isolated from the gland and medium by organic solvent and salt fractionation, trichloroacetic acid precipitation, Sephadex G-100 gel filtrat...

Impact of proteoglycan-4 and parathyroid hormone on articular cartilage.

Proteoglycan-4 (Prg4) protects synovial joints from arthropathic changes by mechanisms that are incompletely understood. Parathyroid hormone (PTH), known for its anabolic actions in bone, increases Prg4 expression and has been reported to inhibit articular cartilage degeneration in arthropathic joints. To investigate the effect of Prg4 and PTH on articular cartilage, 16-week-old Prg4 mutant and...
